Since there is no link between these patients, called acute non-HepA-E hepatitis, and viral hepatitis agents (hepatitis A, B, C, D, and E), the possible etiology, and pathogenesis of this emergency is being investigated. One of the alarming features of this epidemic is the high requirement for liver transplantation in a fraction of the cases. In cases other than hepatitis A, B, C, D, and E, a case definition is made by looking at a series of clinical pictures, including serum transaminase levels and age. As of August 26, 2022, 513 cases of acute hepatitis have been reported in Europe and 1,010 globally. Adenovirus was detected in 75% of cases tested in the UK, but data for other countries are still lacking. The role of other etiologic agents is still under investigation. The exact disease pathogenesis has not yet clear. Evidence of human-to-human transmission of the disease remains unclear. Epidemiological studies are critical in clarifying the uncertainties regarding the existence of links between the cases reported to date. Continuing the national and international surveillance activities of the countries in an organized manner is the most basic issue required for the elimination of the epidemic. Methods: A methodological research. The study was conducted with caregivers of individuals with multiple sclerosis who presented to the neurology clinic of a university hospital between June 2019 and September 2020. The validity and reliability study of the Caregiving Tasks in Multiple Sclerosis Scale (CTiMSS) was performed with a total of 142 individuals, which is more than 5 times the number of items on the scale (24 items). Results: The validity of the CTiMSS was evaluated with language validity, construct validity (CFA), and content validity. The reliability coefficient of the internal consistency analysis (Cronbach's Alpha) was found to be extremely high (0.97). Item-total correlation of the scale was examined, and accordingly, no items were removed from the scale. The scale was administered to 30 patients at a two-week interval to analyze test-retest reliability and time-dependent invariance. Conclusion: The CTiMSS, which was adapted to Turkish society to evaluate the tasks of individuals providing care for individuals with multiple sclerosis, is a valid and reliable tool. We recommend that it should be administered to different groups and cultures to increase its evidence value. CTiMSS is an appropriate measurement tool to determine the duties of caregivers of individuals with multiple sclerosis.Article Brain in Metabolic Syndrome Model: the Effect of Exercises and Caloric Restriction(Marmara Univ, 2022) Alev-Tuzuner, Burcin; Genc-Kahraman, Nevin; Ipekci, Hazal; Ustundag, Unsal Veli; Tunali-Akbay, Tugba; Emekli-Alturfan, Ebru; Yarat, AysenCaloric restriction (CR) and exercise (EX) have impacts on improving metabolic risk factors. This study aimed to investigate the changes in the brain after EX and/or CR in metabolic syndrome (MeS) induced by a high fructose diet in rats. Sprague-Dawley male rats were divided into five groups. Drinking water including 10% fructose solution was given to rats for 12 weeks to develop a MeS rat model. Animals with MeS were submitted to EX and/or CR for 6 weeks. Blood glucose, and brain tissue damage and antioxidant parameters were measured. Brain lipid peroxidation, sialic acid, mucin, fucose levels increased in the MeS group compared to the control (C) group. These parameters reduced significantly in the metabolic syndrome with caloric restriction (MeS+CR) group, and more significantly in the metabolic syndrome with exercise and caloric restriction group (MeS+EXCR), compared to the MeS group. Glutathione levels, superoxide dismutase and catalase activities decreased in the MeS group compared to the C group, increased both in the MeS+CR group, and MeS+EXCR group compared to the MeS group. High fructose diet consumption can lead to brain tissue damage and decreased antioxidant levels were found to be improved best in the MeS+EXCR group.Article Canlı Karaciğer Donör Adaylarında Hepatosteatozun Saptanmasında Biyopsi, Bilgisayarlı Tomografi ve Manyetik Rezonans Görüntülemenin Karşılaştırılması(2021) Sokmen, Bedriye Koyuncu; Şahin, Tolga; Koçak, Erdem; Oral, AlihanAmaç: Karaciğer donöründe hepatosteatoz (HS) varlığı, karaciğer transplantasyonu sonuçları üzerinde olumsuz etkilere sahiptir. Ethanol Extract in the Treatment of Burn Wounds(Marmara Univ, 2022) Erta, Busra; Okuyan, Betul; Sen, Ali; Ercan, Feriha; Onel, Huseyin; Goger, Fatih; Sener, GokselThe overall aim of the present research is to evaluate for the first time the curative effect of Cotinus coggygria leaves on burn injury in an experimental burn model along with its anti-inflammatory and antioxidant activity potential. Also, phenolic compounds of C. coggygria were characterised by LC-MS/MS. Wistar albino rats weighing 200-250 g were exposed to 90 degrees C bath for 10 s to induce burn injury, involving 30% of the total body surface area. In the treatment groups, 5% C. coggygria ethanol extract was applied topically as a cream immediately after the burn. Blood and skin tissue samples were taken after decapitation at the 4th and 48th hours following the burn procedure. Interleukin 1-beta (IL-1 beta) and tumour necrosis factor (TNF-alpha) were determined in serum samples, and hydroxyproline, prostoglandin E2 (PGE2), and myeloperoxidase (MPO) activity and 8-hydroxy-2'-deoxy-guanosine (8-OHdG) levels were determined in skin tissue samples. Increased levels of serum cytokines were decreased with C. coggygria treatment in both periods. MPO activity, prostaglandine (PGE2), and 8-OhdG levels increased, while hydroxyproline levels decreased due to burn damage. On the other hand, these parameters were returned to its normal levels with C. coggygria treatment. In addition, the tissue histology of animals treated with C. coggygria showed a complete epithelialization with increased collagenation. As a result, C. coggygria may be an alternative treatment approach for burns-induced skin damage and wounds.Article Citation - WoS: 7Effect of Flurbiprofen Derivative (sgk597) on Cell Proliferation and Apoptosis of Breast Cancer Cell Lines(Istanbul Univ, Fac Pharmacy, 2022) Atlihan, Irem; Sevinc, Sevgi Kocyigit; Orun, Oya; Yilmaz, Ozgur; Kucukguzel, S. Guniz; Tiber, Pinar MegaBackground and Aims: The incidence of breast cancer is increasing day by day, especially in women. The search for new drugs against breast cancer is the focus of attention in research. Breast cancer and prostate cancer have remarkable biological similarities. Therefore, the 4-(4-chlorophenyl)-3-(1-(2-fluoro-[1,1'-biphenyl]-4-yl)ethyl)-5-((4-fluorobenzyl)thio)-4H-1,2,4-triazole (SGK597) compound that is suppressing cell proliferation in prostate cancer, was studied in MCF-7 breast can-cer and MCF-10A mammary epithelial cell lines. Methods: The WST-8 method was used to determine cell viability and cytotoxicity of SGK597 in MCF-7 and MCF10-A cell lines. The JC-1 test was applied to determine changes in mitochondrial membrane potential. The protein expression levels of Bax, Bcl-2, and c-PARP associated with apoptosis were determined using Western blot analysis. Results: After 24 and 48 hours of incubation of SGK597, the IC50 values were 28.74 pM and 17.28 pM for MCF-7; 65.9 pM and 50.5 pM for MCF-10A, respectively. Mitochondrial membrane potential showed a tendency toward depolarization in MCF-7 cells as a result of increasing concentration of SGK597, while the same tendency was not seen for MCF-10A. As a result of western blot experiments, no increase in the Bax/Bcl-2 ratio and c-PARP expression level was observed, indicating no apoptosis. Conclusion: It was observed that the compound SGK597 suppressed MCF-7 cell proliferation. These results indicate that SGK597 may be a candidate compound for use as an anticancer agent. Prior studies have identified PDI is highly expressed in many different cancer types and presented as a new potential target for cancer treatment. Here, we investigated vitamin D and its analogue paricalcitol in silico interaction of the human PDI and inhibition of PDI reductase activity in vitro. We observed a non-covalent mechanism where the main skeleton of the vitamin D3 ans paricalcitol sturcture is located at the hydrophobic site in the b' domain of PDI and forms a hydrogen bond with a residue (His138) in tihs domain. They also form multiple weak hydrophobic interactions with various chemical groups of the b' subunit. For the first time, we demonstrate that 1,25-dihydroxyvitamin D3 (1a,25(OH)2 vitamin D3) and paricalcitol inhibit the PDI reductase activity in vitro and their IC50 values are 20.79±1.43 nmol/L and 32.83±3.15 nmol/L respectively. The two compounds can also block the denistrosation activity of PDI.Article The Effect on Birth Pain and Process of the Freedom of Movement in the First Stage of Labor: a Randomized Controlled Study(2022) Yıldız, Hatice; Karaman, Özen EsraObjective: To determine the effect of freedom of movement implemented in the dilatation stage, which is the first stage of labor, on the labor process in pregnant women who will give their first birth. Extract and Apocynin on Lens Oxidative Damage and Boron Levels in Rats With a High Fat-Diet(Galenos Publ House, 2021) Yasar, Ruya Kuru; Kuru, Dilruba; Sen, Ali; Sener, Goksel; Ercan, Feriha; Yarat, AysenObjectives: Nutritional obesity causes oxidant damage in the body and cataract formation in the lenses by increasing the formation of free radicals. Myrtus communis leaf extracts (Myr) have antioxidant properties, and apocynin (Apo) is an effective NADPH-oxidase inhibitor. The data on tissue boron levels are quite lacking. The aim of this novel study was to investigate the effects of Myr and Apo treatment on boron levels and oxidative lens damage in rats fed a high-fat diet (HFD). Materials and Methods: Wistar albino male rats were randomly divided into four groups: the control group, HFD group, HFD + Myr group, and HFD + Apo group. Body weight and blood lipids were determined before and after the experiment. After decapitating the rats, the lenses were removed and homogenized. Catalase (CAT) and superoxide dismutase (SOD) activities and boron, malondialdehyde (MDA), and reduced glutathione (GSH) levels in the lens homogenates were determined. Results: The HFD increased serum triglyceride (p<0.05), total cholesterol level (p<0.001), body weight (p<0.001), and lens MDA levels (p<0.01) and decreased lens GSH (p<0.05) and boron level (p<0.01), SOD (p<0.001), and CAT activity (p<0.001). However, Myr and Apo treatment reduced the rats' body weight (p<0.001), serum triglyceride (p<0.05), and total cholesterol level (p<0.001) and increased lens boron (p<0.01; p<0.001), GSH levels (p<0.05; p<0.01), and CAT activity (p<0.001). Conclusion: Both Myr and Apo may be able to reduce oxidative stress in the lenses of obese rats caused by HFD by increasing boron levels.
