Article 19. Article Citation - WoS: 1Citation - Scopus: 1A Comparative Study of Biochemical, Antimicrobial Effects and Phytochemical Composition Analysis of Glycyrrhiza Glabra L. Since there is no link between these patients, called acute non-HepA-E hepatitis, and viral hepatitis agents (hepatitis A, B, C, D, and E), the possible etiology, and pathogenesis of this emergency is being investigated. One of the alarming features of this epidemic is the high requirement for liver transplantation in a fraction of the cases. In cases other than hepatitis A, B, C, D, and E, a case definition is made by looking at a series of clinical pictures, including serum transaminase levels and age. As of August 26, 2022, 513 cases of acute hepatitis have been reported in Europe and 1,010 globally. Adenovirus was detected in 75% of cases tested in the UK, but data for other countries are still lacking. The role of other etiologic agents is still under investigation. The exact disease pathogenesis has not yet clear. Evidence of human-to-human transmission of the disease remains unclear. Epidemiological studies are critical in clarifying the uncertainties regarding the existence of links between the cases reported to date. Continuing the national and international surveillance activities of the countries in an organized manner is the most basic issue required for the elimination of the epidemic. Extract Ameliorates Liver Fibrosis in Bile Duct-Ligated Rats(Istanbul University Press, 2022) Albayrak, O.; Ozbeyli, D.; Sen, A.; Cevik, O.; Erdogan, O.; Ercan, F.; Sener, G.Objective: The aim is to examine the possible protective effect of Cuscuta sp. extract against liver damage induced by biliary obstruction in rats. Materials and Methods: To induce biliary obstruction, the bile duct ligation (BDL) method was used. Sprague Dawley rats were allocated to 4 groups: Control (C), Cuscuta (CUS), bile duct ligation (BDL), and bile duct ligation + Cuscuta (BDL+CUS). Control and BDL rats were given physiological saline (SF), while CUS and BDL+CUS groups were administered 250 mg/ kg of Cuscuta extract by oral gavage. At the end of 28th day, the rats were decapitated, serum and tissue samples were collected, and aspartate aminotransferase (AST), alanine transaminase (ALT), and direct and total bilirubin (DB and TB) levels were determined in blood samples. In liver tissues, transforming growth factor-β (TGF-β), 8-hydroxyguanosine (8-OHdG), hydroxyproline, and sodium-potassium ATPase (Na+/K+-ATPase) levels were determined. Results: Serum samples of rats with cholestasis had high ALT, AST, DB, and TB levels, while TGF-β, 8-OHdG, and hydroxyproline concentrations were found to be significantly high in tissues. Hepatic Na+/K+-ATPase levels were decreased through biliary obstruction. Biochemical parameters were drastically reversed by Cuscuta care; also, this was supported histologically. Conclusion: Results showed that Cuscuta extract, through its antioxidant and anti-inflammatory properties, provided protection against oxidative injury by biliary obstruction. Also, these results confirm the traditional use of Cuscuta sp. as hepatoprotective. © Ankara Medical Journal. All rights reserved.Article Citation - WoS: 10Citation - Scopus: 11The Effect of cotinus Coggygria L. Ethanol Extract in the Treatment of Burn Wounds(Marmara Univ, 2022) Erta, Busra; Okuyan, Betul; Sen, Ali; Ercan, Feriha; Onel, Huseyin; Goger, Fatih; Sener, GokselThe overall aim of the present research is to evaluate for the first time the curative effect of Cotinus coggygria leaves on burn injury in an experimental burn model along with its anti-inflammatory and antioxidant activity potential. Also, phenolic compounds of C. coggygria were characterised by LC-MS/MS. Wistar albino rats weighing 200-250 g were exposed to 90 degrees C bath for 10 s to induce burn injury, involving 30% of the total body surface area. In the treatment groups, 5% C. coggygria ethanol extract was applied topically as a cream immediately after the burn. Blood and skin tissue samples were taken after decapitation at the 4th and 48th hours following the burn procedure. Interleukin 1-beta (IL-1 beta) and tumour necrosis factor (TNF-alpha) were determined in serum samples, and hydroxyproline, prostoglandin E2 (PGE2), and myeloperoxidase (MPO) activity and 8-hydroxy-2'-deoxy-guanosine (8-OHdG) levels were determined in skin tissue samples. Increased levels of serum cytokines were decreased with C. coggygria treatment in both periods. MPO activity, prostaglandine (PGE2), and 8-OhdG levels increased, while hydroxyproline levels decreased due to burn damage. On the other hand, these parameters were returned to its normal levels with C. coggygria treatment. In addition, the tissue histology of animals treated with C. coggygria showed a complete epithelialization with increased collagenation. As a result, C. coggygria may be an alternative treatment approach for burns-induced skin damage and wounds.Article The Effect of Vitamin D and Paricalcitol on Protein Disulfide Isomerase(Marmara University, 2025) Koksal, Murat; Şekerler, Turgut; Şener, AzizeProtein disulfide isomerase (PDI), a multifunctional protein plays an important role as oxidoreductase, isomerase and chaperone in the cell. Prior studies have identified PDI is highly expressed in many different cancer types and presented as a new potential target for cancer treatment. Here, we investigated vitamin D and its analogue paricalcitol in silico interaction of the human PDI and inhibition of PDI reductase activity in vitro. We observed a non-covalent mechanism where the main skeleton of the vitamin D3 ans paricalcitol sturcture is located at the hydrophobic site in the b' domain of PDI and forms a hydrogen bond with a residue (His138) in tihs domain. They also form multiple weak hydrophobic interactions with various chemical groups of the b' subunit. For the first time, we demonstrate that 1,25-dihydroxyvitamin D3 (1a,25(OH)2 vitamin D3) and paricalcitol inhibit the PDI reductase activity in vitro and their IC50 values are 20.79±1.43 nmol/L and 32.83±3.15 nmol/L respectively. The two compounds can also block the denistrosation activity of PDI.Article The Effects Of Panax Ginseng on Serum Oxidative Stress Following Bisphenol a Exposure(Istanbul Univ, 2024) Fazalyar, Najiullah; Pazarbasi, Seren Ede; Dorucu, Dogancan; Sener, Goksel; Tunali-Akbay, TugbaObjective: Bisphenol A (BPA) is a toxic compound that causes oxidative stress by disrupting antioxidant enzymes and promoting tissue lipid peroxidation. This study aimed to examine the impacts of BPA on serum oxidative stress in rats and to detect the antioxidant feature of Panax ginseng (PxG) in reducing BPA-induced oxidative stress. Materials and Methods: Wistar Albino rats (250-300 g) were divided into control, control + PxG, BPA, and BPA + PxG groups. 50 mg/kg BPA and 100 mg/g PxG were given for six weeks. Serum total antioxidant and oxidant status, lipid peroxidation, and glutathione levels were determined. Results: BPA administration increased total oxidant status and lipid peroxidation, while PxG administration to the BPA group decreased these parameters. PxG also increased total antioxidant status and glutathione levels compared to the BPA group. Conclusion: BPA was seen to cause an increase in oxidative parameters and PxG administration to restore the oxidative stress that was generated after BPA exposure, suggesting that this may help to prevent the adverse effects caused by BPA exposure.Article Ensemble-Based Alzheimer's Disease Classification Using Features Extracted From Hog Descriptor and Pre-Trained Models(Sakarya University, 2024) Muzoğlu, Nedim; Akbacak, EnverAlzheimer's Disease is the most common type of dementia and is a progressive, neurodegenerative disease. The disease worsens over time, and the patient becomes bedridden, unable to move or understand what is happening around him. The main concern of medicine is to slow down the progression of the disease for which no treatment has yet been developed. Artificial intelligence studies have achieved significant success in detecting many diseases. In this study, an artificial intelligence-based approach that uses MR images of the early stage of Alzheimer's Disease to detect the disease at an early stage is presented. Initially, a new dataset was created through the application of the fuzzy technique, thereby expanding the feature space. Then, an ensemble learning-based hybrid deep learning model was developed to reduce the misclassification rate for all classes. The features derived from the inception module, residual modules, and histogram of oriented gradients descriptor are subjected to classification through bagging and boosting algorithms. The proposed model has surpassed many state-of-the-art studies by achieving a high success rate of 99.60% in detecting Alzheimer's disease in its early stages.Article Farklı Kilo Verme Yöntemlerinin ve Yöntemlere Uyumun Kilo Verme ve Kontrolü ile İlişkisi(2021) Çıtıl, Rıza; Eğri, Mücahit; Gurdal, Osman; Kişioğlu, Ahmet Nesimi; İlhan, İlter; Gültekin, Fatih; Orhan, HikmetAmaç: Obezite, çağımızın en önemli sağlık sorunlarından biridir. Var. Cicla Improves Memory Deficits in Intracerebroventricular Streptozotocin Injected Rats: Role on Neuroinflammation(Marmara Univ, 2021) Ertas, Busra; Topal, Fadime; Gulhan, Rezzan; Yanardag, Refiye; Sacan, Ozlem; Sener, GokselAlzheimer's disease is a challenging disease for patients due to progressive loss of cognition and behavioral disorders. Disruption of cholinergic transmission and neuroinflammation are the most important mechanisms underlying cognitive damage. Beta vulgaris L. var. cicla (BV) has been reported to have various pharmacological effects associated with its rich antioxidant content. In addition, anti-cholinesterase and antiinflammatory activities of BV have been demonstrated in vitro. The aim of this study is to elucidate the therapeutic effect of BV against cognitive impairment, reduction in cholinergic transmission and neuroinflammation caused by intracerebroventricular (ICV) administration of streptozotocin (STZ). STZ was administered bilaterally at a dose of 3 mg/kg via ICV to rats, and BV treatment at a dose of 2 g/kg for 21 days was administered orally to STZ-induced animals. After behavioral tests, AChE activity, TNF-alpha and IL-1 beta levels were measured in hippocampus and cortex tissues excised from decapitated animals. Novel object recognition and passive avoidance test showed that the treatment of BV reverted the ICV-STZ induced memory dysfunctions in rats. Furthermore, increased AChE levels in the hippocampal and cortical tissues of STZ-induced rats were significantly reduced with 21 days of BV treatment. In conclusion, these results confirm that STZ administration caused cholinergic hypofunction, neuronal inflammation and cognitive dysfunction in rats, and BV therapy significantly inhibited these changes with its potential neuroprotective activity.Article In Vitro Efficacy of Sumac (Rhus coriaria) Extracts Against Leishmania Tropicana and Leishmania Mexicana: A Preliminary Study from Turkiye(Istanbul Univ, 2025) Mete, Ergun; Ozel, Yener; Bardakci, Hilal; Durmuskahya, Cenk; Koseler, Aylin; Kurt, OzgurObjective: Cutaneous leishmaniasis (CL) is a common clinical manifestation of leishmaniasis. Here, the in vitro anti-leishmanial efficacy of sumac extracts was tested for the first time on both Leishmania (L.) tropica and L. mexicana isolates using Rhus (R.) coriaria plant, which was collected in western Anatolia. Materials and Methods: The dried and powdered fruits of R. coriaria were macerated in acetone, ethyl alcohol, and ethyl alcohol-water mixture at room temperature for two days. The pooled extracts were evaporated under reduced pressure and lyophilized form for the study. Isolates of L. tropica and L. mexicana in Acibadem University R&D Laboratory were initially thawed and cultivated in NNN medium. Assessments were made using the haemocytometer and MTT methods at 24 and 48 h, compared with meglumine antimoniate as the control group. Results: For L. tropica, the effective concentration ranges of the extracts and the infusion were found to be 578.13-289.06 pg/mL and 289.06-144.53 pg/mL, respectively. For L. mexicana, the ranges were found to be 289.06-144.53 pg/mL and 144.53-72.27 pg/mL, respectively. It was shown that all extracts of R. coriaria were effective against both L. tropica and L. mexicana in higher doses, compared to meglumine antimoniate. Conclusion: An interesting finding was that higher sumac doses were required to eliminate L. tropica of the Old World, compared to L. mexicana of the New World. In addition, the aqueous alcohol extract showed efficacy that lasted for 48 h in half doses compared to others in L. tropica. Further assessments for both the identification of the active compounds within R. coriaria and their efficacy in vivo are planned.Article Citation - WoS: 3Citation - Scopus: 3Investigation of the Protective and Therapeutic Efficacy of myrtus Communis Extract in Aluminum Chloride and Dgalactose-Induced Alzheimer's Disease in Rats(Marmara Univ, 2022) Yalman, Kiibra; Sen, Ali; Cevik, Ozge; Kadioclu-Yaman, Beril; Ertas, Biiva; Yildiz, Sila; Sener, GokselThis study investigated the possible protective and therapeutic effects of Myrtus communis subsp. communis ethanol extract (MC) in aluminum chloride (AlCl3) and D-galactose (D-Gal) induced Alzheimer's disease in rats. MC was orally given to rats as a protective treatment for 90 days and, in other two groups starting from the 60th day MC (100-200 mg/kg) was administered, concomitantly with AlCl3 and D-Gal. Learning and memory functions were evaluated by the behavioral tests. Biological activities of MC treatment were examined in hippocampal tissues by ELISA tests. D-Gal and AlCl3-treated rats showed increased amyloid beta (A ss) and 8-hydroxy-2-deoxyguanosine (8OHdG) levels, acetylcholinesterase activity and decreased neprilysin, Na+-K+ATPase and SOD levels in parallel with a decrease in Novel Object Recognition Test, Morris Water Maze and Passive Avoidance Test scores. On the other hand, MC administration reversed the behavioral impairments and improved learning and memory. Moreover, MC treatment decreased A ss and 8-OHdG levels and acetylcholinesterase activity and increased neprilysin levels, Na+-K+ATPase and SOD levels. Our results suggest that MC has beneficial effects on cognitive and neuronal functions through its anticholinesterase and antioxidant properties.
