Article Sub-Cytotoxic Mitochondrial Stress in Cardiomyocytes and Whole-Organism Toxicity in C. Elegans Induced by Molnupiravir(SAGE Publications Inc, 2026) Gunaydin-Akyildiz, Aysenur; Keskin, Zehra; Ozhan, Gul; Kuran, Ebru Didem; Gulec, MeltemMany antiviral agents are known to induce off-target mitochondrial toxicity due to the prokaryotic origin of mitochondria. Mitochondrial dysfunction is frequently linked to cardiotoxicity. We aimed to elucidate the mitochondrial toxicity profile of molnupiravir via focusing on mitochondrial dynamics, biogenesis, and oxidative stress in cardiac cells. Mitochondrial function was evaluated by luminometric measurement of ATP (adenosine triphosphate) content, and flow cytometric analysis of mitochondrial membrane potential, and mitochondrial mass. The expression levels of genes involved in mitochondrial fusion-fission were assessed by RT-PCR. In addition, molecular docking analysis was performed to evaluate the interaction between molnupiravir and the dynamin related-protein DRP1. Protein carbonylation was determined as an oxidative stress parameter. Toxicity evaluation was further investigated in Caenorhabditis elegans to support the in vitro findings at the organismal level. Molnupiravir exposure led to a significant dose-dependent reduction in intracellular ATP level and mitochondrial mass, accompanied by increased protein carbonylation. Mitochondrial membrane potential remained slightly increased. Alterations in the expression of genes regulating mitochondrial dynamics suggested an imbalance between fusion and fission processes, while mitochondrial biogenesis-related signaling was progressively suppressed. C. elegans exposed to higher concentrations of the drug (20-500 & micro;M) exhibited significant lifespan reduction at all doses. Molecular docking analysis demonstrated a moderate binding affinity of molnupiravir to DRP1, supporting a potential direct interaction with mitochondrial fission machinery. In conclusion, our results demonstrate that molnupiravir induces mitochondrial stress through oxidative damage, impaired biogenesis, and altered dynamics, emphasizing the need for careful evaluation of mitochondrial safety of molnupiravir in cardiac tissue.Article Safety and Efficacy of Colchicine in COVID-19 Treatment: Systematic Review and Meta-Analysis(Elsevier, 2025) Nada, Ahmed Hosney; Ibrahim, Ismail A.; Asar, Nada Khalid; Qenawy, Abdulrahman; Mohammed, Mariam M.; Wagdy, Mohamed; Farouk, Heidi SherifBackground: Colchicine is an anti-inflammatory drug used for the treatment of gout and other autoinflammatory conditions. Several trials reported promising results of the efficacy of colchicine in Covid-19 due to its antiinflammatory properties. However, applying these results to clinical settings remains the subject of ongoing research. Methods: We performed a systematic review and meta-analysis on the efficacy and safety of colchicine in the treatment of Covid-19. PubMed, Cochrane Library, Web of Science, and Scopus were searched for relevant studies. The primary outcomes were 28-day mortality, invasive mechanical ventilation, non-invasive mechanical ventilation, and ICU admissions. The risk ratio was used to compare effectiveness between the two groups. Subgroup analysis was done for C-reactive protein and duration of hospitalization. Results: 17 randomized controlled trials with a total of 25478 patients were included. The overall Risk ratio didn't favor any of the two groups in terms of 28-day mortality (RR =1.03, 95 % CI [0.93:1.15], P = 0.58), noninvasive mechanical ventilation (RR = 0.81, 95 % CI [0.44:1.48], P = 0.49), ICU admission (RR = 0.89, 95 % CI [0.56:1.41], P = 0.62). The overall mean difference (MD) did not show statistical significance between both groups in terms of Creactive protein (CRP) (mg/dl) (MD = -1.21, 95 % CI [-2.42:0.01], P = 0.05), lactate dehydrogenase (LDH) (U/ L) (MD = 50.95, 95 % CI [-92.07: 193.98], Ferritin (ng/ml) (MD = 128.08, 95 % CI [51.97:204.18], P = 0.001), ICU length of stay (MD = -0.09, 95 % CI [-0.34:0.15], P = 0.45) and duration of hospitalization (MD = -0.41, 95 % CI [-1.56:0.73], P = 0.48). Conclusion: In Covid-19 treatment, colchicine didn't result in significant benefits in terms of clinical outcomes. More large-scale randomized clinical trials with standardized dosages and long-term follow-up are needed for further investigation into the colchicine effect.Article How Have Occupational Therapy Services Been Delivered During the COVID-19 Pandemic(Nova Southeastern Univ., 2024) Yucel, Hulya; Aydoner, SelenThe provision of traditional healthcare services has encountered obstacles due to the Coronavirus Disease-2019 (COVID-19) pandemic. The inability of healthcare professionals and clients to survive the pandemic and the inadequacy of personal protective equipment have created uncertainties regarding the appropriate practices for addressing individuals' health conditions. Furthermore, the field of rehabilitation, including occupational therapy, has experienced changes in the delivery of traditional practices. Therefore, this review aimed to examine occupational therapy services delivered during the COVID-19 pandemic. As a result of the literature review, the most commonly provided services by occupational therapists during COVID-19 were discussed and a total of 87 references were accessed. These services were grouped under the headings of acute/intensive care unit, post-COVID-19, telerehabilitation, preventive mental health, school-based occupational therapy, occupational therapy education, and ergonomics. It was seen that occupational therapists had several challenges during the pandemic. They had to adapt their interventions according to the new conditions and so they have frequently used technology. It is crucial for the effectiveness of the rehabilitation that occupational therapy services continue in challenging and unexpected situations such as the COVID-19 pandemic. It is necessary to strengthen future occupational therapy practices based on standards through collaboration among occupational therapists. It is believed that this review will shed light on the role and importance of occupational therapy during the challenging COVID-19 pandemic.Article Perceived Stress and Performance of Daily Activities Among Emergency Healthcare Workers During the COVID-19 Pandemic(Sage Publications Ltd, 2025) Yücel, Hülya; Bektas, Selen AydonerBackground: During the COVID-19 pandemic, healthcare workers were exposed to intense stress in the emergency departments and sudden changes in their daily lives. Objective: The study aimed to investigate the impact of the pandemic on performance and participation in the activities of daily living, personal lives, and overall well-being of the healthcare workers in an emergency department. Methods: Seventy-eight people working in an emergency department of a university hospital (100% of those eligible) participated in the study during June-July 2022. A self-report survey, cross-sectional design was used to collect data. Participants completed the Perceived Stress Scale-10 (PSS) and the Canadian Occupational Performance Measure (COPM). Independent sample t-tests, Pearson correlation, and multiple regression analysis were used for statistical analysis. Results: There was a significant negative correlation between PSS and COPM Performance (r = −0.524) as well as between PSS and COPM Satisfaction (r = −0.508), indicating that higher levels of stress were associated with lower performance and satisfaction. In addition, work-related factors, including working experience, daily working hours, and time available for rest at work, were significantly associated with PSS, COPM Performance, and Satisfaction (p < 0.05). The strongest multivariate associations (p = 0.001) were found between lacking time to rest at work and PSS (direct) and COPM Performance and Satisfaction (inverse). Conclusion: The stress experienced by emergency healthcare workers during COVID-19 negatively impacted their performance, daily activities, personal lives, and well-being. A potential implication is that policy-makers addressing work-related factors and implementing regulations could improve the well-being of these workers in their professional practices. © 2025 Elsevier B.V., All rights reserved.
