    In Vivo Antioxidant and Anti-Inflammatory Effects of Myrtus Communis Against Ionizing Radiation-Induced Gastrointestinal Injury: Trod-Grog Study

    OBJECTIVE: This study aimed to investigate the in vivo radioprotective effects of Myrtus communis (MC) on the gastrointestinal system. METHODS: A total of 30 female rats were divided into four groups: i) Control; ii) irradiation (IR) only; iii) MC-pretreated; and iv) MC-treated. The rats received oral MC extract (100 mg/kg/day) for 4 days before exposure to 10 Gy IR or continued until sacrifice. On the fourth day of IR exposure, the rats were sacrificed, and histopathological and biochemical analyses were performed on the ileum, pancreas, and liver tissues. RESULTS: Malondialdehyde and myeloperoxidase levels decreased in both MC-treated groups, while glutathione levels and Na+-K+-ATPase activity increased (p<0.01), with significant histopathological improvements compared to the IR-only group. CONCLUSION: The results of this study demonstrate that MC significantly decreases ionizing radiation-induced oxidative and inflammatory damage in the gastrointestinal systems of rats. Therefore, it may be regarded as a new candidate with radioprotective potential for future clinical application.

    Myrtus Communis Ameliorates Ionizing Radiation-Induced Cardiopulmonary Injury in Rats: Trod-Grog Study

    Objectives: Ionizing radiation (IR), widely used in diagnostic and therapeutic procedures, can damage vital organs such as the heart and lungs through oxidative stress. This study aims to assess the potential radioprotective effect of Myrtus communis (MC) against cardiopulmonary injury. Methods: Thirty female rats were divided into four groups. Control (C) and IR (R) groups received oral saline. The treatment (R+MC) and pretreatment (R+preMC) groups received MC (100 mg/kg) for 4 days (starting on the day of IR) and 8 days (starting 4 days before IR), respectively. All IR-exposed groups (R, R+MC, R+preMC) received a single 10 Gy whole-body irradiation. Histopathological changes were assessed by hematoxylin-eosin staining, while oxidative stress was evaluated by measuring malondialdehyde (MDA), glutathione (GSH), myeloperoxidase (MPO), nitric oxide (NO), superoxide dismutase (SOD), glutathione S-transferase (GST), and tissue factor activity (TFa) levels. Protein profiles in tissues were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Results: Histopathologically, MC reduced alveolar and cardiomyocyte damage in both R+MC and R+preMC groups. IR increased all oxidative stress markers and decreased antioxidant parameters in heart and lung tissues (p < 0.05-0.001). Both MC treatment and pretreatment reversed these effects, significantly reducing oxidative/inflammatory markers and restoring antioxidant enzyme activities (p < 0.05-0.001). The R+preMC group demonstrated a stronger protective effect than the R+MC group. Conclusion: Our study shows that MC has a radioprotective effect on the cardiopulmonary system by decreasing oxidative damage. MC appears to be a promising natural compound for advanced radioprotection research, and further molecular and clinical studies could clarify its mechanisms and potential applications<bold>.</bold>

    The Therapeutic Role of Ginseng in Promoting Hippocampal Neurogenesis and Ameliorating Cognitive Function Following Whole Brain Radiotherapy in Rats

    Whole-brain radiotherapy (WBRT) is a prevalent technique for managing multiple intracranial metastases, however, the cognitive damage in long-term survivors due to WBTR is a critical concern that impacts patients' quality of life. Panax ginseng, a bioactive compound recognized for its neuroprotective benefits, also enhances cognitive functions, including memory and learning. This study aims to examine the potential protective effects of Panax ginseng supplementation on cognitive dysfunction and the levels of neurogenesis-related proteins in the hippocampus of rats that underwent WBRT, which was delivered as 3 fractions of 6 Gy (total dose 18 Gy) using a linear accelerator. Thirty-six male Sprague-Dawley rats were divided into three groups: radiation, ginseng treatment, and control. After 60 days of Panax ginseng administration (100 mg/kg), behavior tests (Morris water maze and novel object recognition) were performed, followed by western blot analysis of the hippocampus. Results indicated that Panax ginseng supplementation ameliorated radiation-induced cognitive impairments. Additionally, western blot analyses revealed that Panax ginseng promoted neuronal recovery and neuroplasticity processes in the hippocampus, simultaneously exhibiting a neuroprotective mechanism by reducing apoptosis and neurotoxicity markers. Panax ginseng ameliorates cognitive dysfunction after WBRT by enhancing neurogenesis and diminishing cell death in the hippocampus.

    NADPH Oxidase-2 Inhibitor Apocynin Attenuates High-Fat Diet-Induced Kidney and Bladder Injury

    Objective: This study aimed to evaluate the effects of NADPH oxidase-2 (NOX-2) inhibitor apocynin (APC) on high-fat diet (HFD)induced renal and bladder injury. Materials and Methods: Wistar albino rats were divided into 4 groups: Control, HFD, HFD+dimetyl sulfoxide (DMSO), and HFD+APC. Rats in HFD, HFD+DMSO, and HFD+APC groups were fed with HFD for sixteen weeks. In the last 4 weeks of the experiment, either DMSO or APC (25 mg/kg, dissolved in DMSO) was applied to the HFD+DMSO or HFD+APC groups. Lipid profiles and leptin values were measured in blood serum. Renal and bladder oxidant/antioxidant parameters, histological changes in the tissues, NOX-2-, nuclear factor kappa B (NF-& kgreen;B)-immunopositive and apoptotic cells were evaluated. Results: At the end of the experiment, leptin, cholesterol, and triglyceride levels were higher and high-density lipoprotein levels were lower in the HFD and HFD+DMSO groups compared to controls. In these experimental groups, an increase in malondialdehyde, 8-hydroxy-deoxyguanosine and myeloperoxidase levels and a decrease in glutathione levels, as well as an increase in collagen, NOX-2-and NF & kgreen;-B-immunopositive and apoptotic cells were found. Also, a deterioration in kidney and bladder morphology was observed. All these biochemical and histopathological findings improved in the HFD+APC group. Conclusion: High-fed diet causes renal and bladder injury by increasing NOX-2 activity and inflammation via oxidative stress. APC might alleviate tissue injury by inhibiting oxidative stress.

    The Effect of <i>myrtus Communis</I> L. Extract on Nephrolithiasis Model in Rats

    OBJECTIVE: Nephrolithiasis is a common urological disease that can lead to renal failure. Oxidative stress has been shown to be a contributing factor for nephrolithiasis and many agents have been studied to prevent and treat oxidative stress-related nephrolithiasis and renal damage. Myrtus communis (MC) extract has been shown to be an important antioxidant in different animal models. In this study, MC extract was administered preventively or therapeutically to rats with kidney stones, and its effectiveness was investigated. METHODS: Wistar albino rats were divided into four groups (n=8); control (C), ethylene glycol (EG), EG+preventive MC, and EG+curative MC groups. The nephrolithiasis model was created by adding 0.75% EG to drinking water for 8 weeks. Ultimately, 24-hour urine was collected to measure calcium, citrate, and creatinine levels. After decapitation, kidney tissues were harvested for histological analyses, measurement of osteopontin and 8-hydroxydeoxyguanosine (8-OHdG) levels, and N-acetyl-beta-glucosaminidase (NAG), myeloperoxidase (MPO) and caspase-3 activities. RESULTS: In 24-hour urine samples, calcium, citrate and creatinine levels were decreased in the EG group, while oxalate levels were increased and in treatment groups these parameters returned to control levels. MPO, 8-OHdG, caspase-3 and NAG activity were significantly increased in tissue and these changes were reversed in both MC groups. Histological findings also supported the biochemical parameters. CONCLUSION: MC can reduce oxidative stress and histopathological changes in kidney tissues in rat nephrolithiasis model when used as either a preventive or therapeutic agent. If supported with further clinical trials, MC might have clinical implications in preventing oxidative renal cell injury and ultimately kidney stone formation.

    Apocynin Ameliorates Testicular Toxicity in High-Fat Diet-Fed Rats by Regulating Oxidative Stress

    Objective: The purpose of this study was to examine the effects of apocynin (APC), an inhibitor of NADPH oxidase (NOX), on high-fat diet (HF)induced testis cytotoxicity.Methods: Wistar albino rats were divided into three groups as control, HF and HF+APC groups. Rats in HF and HF+APC groups were fed using HF for 16 weeks and in the last four weeks of this period vehicle solution or APC (25 mg/kg) was administered orally five days a week, respectively. Control group was fed with standart lab chow for 16 weeks. Cholesterol, triglyceride, high-density lipoproteins, leptin, estrogen, testosterone, LH and FSH were estimated in blood serum. Sperm parameters were analysed from the epididymis. Testicular malondialdehyde, 8-hydroxy-2deoxyguanosine, glutathione, superoxide dismutase and myeloperoxidase levels were estimated biochemically. Testicular morphology, proliferative, apoptotic and NOX2-positive cells were analysed histologically.Results: HF-induced obesity caused significant alterations in serum lipid and hormone profiles. Testicular malondialdehyde, 8-hydroxy-2deoxyguanosine, and myeloperoxidase levels increased, glutathione and superoxide dismutase levels decreased in this group. Moreover, altered sperm parameters, increased degenerated seminiferous tubules, apoptotic and NOX2 - positive cells and decreased proliferative cells were observed in the HF group. All these biochemical and histological alterations improved in the HF+APC group.Conclusion: HF-induced obesity causes altreations in lipid values, sperm parameters and testicular morphology by increasing oxidative stress through NOX2 activity. Apocynin might prevent testis damage via regulating oxidant/antioxidant balance.

    <i>cotinus Coggygria</I> Scop. Attenuates Acetic Acid-Induced Colitis in Rats by Regulation of Inflammatory Mediators

    In traditional medicine, many medicinal plants are used in the treatment of various diseases caused by inflammation. The objective of the present study is to elucidate for the first time the effects of Cotinus coggygria (CC) ethanol extract (CCE) on colonic structure and inflammation of acetic acid-induced ulcerative colitis in rats. Colonic damage was assessed using disease activity index score, enzyme-linked immunosorbent assay, and hematoxylin-eosin staining. Also, in vitro antioxidant activity of CCE was investigated by ABTS methods. Total phytochemical content of CCE was measured spectroscopically. Acetic acid caused colonic damage according to disease activity index and macroscopic scoring. CCE significantly reversed these damages. While the levels of proinflammatory cytokines TNF-alpha, IL-1beta, IL-6, and TGF-1beta increased in tissue with UC, IL-10 level decreased. CCE increased inflammatory cytokine levels to values close to the sham group. At the same time, while markers indicating disease severity such as VEGF, COX-2, PGE2, and 8-OHdG indicated the disease in the colitis group, these values returned to normal with CCE. Histological research results support biochemical analysis. CCE exhibited significant antioxidant against ABTS radical. Also, CCE was found to have a high content of total polyphenolic compounds. These findings provide evidence that CCE might be benefit as a promising novel therapy in the treatment of UC in humans due to high polyphenol content and justify the use of CC in folkloric medicine for treatment of inflamed diseases.

    Carvacrol Improves Cognitive Dysfunction by Decreasing Amyloid-Β Accumulation and Regulating Neuroinflammation in Ovariectomized Renovascular Hypertensive Rats

    Hypertension contributes to both the development and progression of brain damage and cognitive dysfunction in the postmenopausal period in women. Carvacrol (CAR), which can easily cross the blood-brain barrier, exhibits neuroprotective properties due to its antioxidant, anti-inflammatory, and anti-apoptotic effects. In the present study, we have examined the effect of CAR treatment on learning-memory impairment in a post-menopausal hypertensive rat model that was induced by ovariectomy following two-kidney, one-clip renovascular hypertension surgery. From the third week after the establishment of renovascular hypertension in ovariectomized rats, CAR (40 mg/kg) was administered once daily for consecutive 7 weeks by gastric gavage. Systolic blood pressure was estimated by the tail-cuff method once a week. At the end of the study, cognitive functions were evaluated with behavioral tests and also neurochemical changes were measured in serum, cortex, and hippocampus by ELISA test. Blood pressure was decreased with CAR treatment in hypertensive rats. Serum estrogen levels decreased in ovariectomized rats and did not change with CAR treatment. CAR demonstrated beneficial effects on learning and memory tests as determined by increased recognition index, the number of platforms crossed, and time spent in the target quadrant. Due to CAR treatment, there was a marked reduction in the hippocampal and cortex amyloid-beta, osteopontin, interleukin-6 and tumor necrosis factor-alpha levels, and acetylcholinesterase activity, while an increment in neprilysin and interleukin-10 levels was found. In conclusion, since CAR suppressed amyloid-beta deposition and neuroinflammation in ovariectomized-hypertensive rats, it is thought that it may be protective against memory disorders in postmenopausal hypertensive women.

    Apocynin Exhibits an Ameliorative Effect on Endothelial Dysfunction/ Atherosclerosis-Related Factors in High-Fat Diet-Induced Obesity in Rats

    Objective: The aim of this study was to reveal the effect of apocynin (APO) on the factors involved in obesity-related endothelial dysfunction (ED) and atherosclerosis (AS). Materials and Methods: Male Wistar albino rats were divided into control (CNT), high-fat diet (HFD) and HFD+APO groups. HFD and HFD+APO groups were fed HFD for sixteen weeks. APO (25 mg/kg) was administered to the HFD+APO group for the last four weeks. The effects of APO on: AS-related metabolic parameters (triglyceride, total cholesterol, high-density lipoprotein-cholesterol, insulin and leptin), oxidative stress (OS), [ malondialdehyde, glutathione, nicotinamide adenine dinucleotide phosphate (NADPH)oxidase-2, oxidised-low-density lipoprotein (ox-LDL) and 8-hydroxy-2-deoxyguanosine], low-density lipoprotein and ox-LDL uptake potential (activin receptor-like kinase-1 and lectin-like oxidized low-density lipoprotein receptor-1, respectively), tissue inflammation inducible-nitric oxide synthase, nitric oxide), programmed cell death (terminal deoxynucleotidyl-transferase-dUTP-nick-end (alpha-smooth muscle actin), histology and ultrastructure of thoracic aorta were evaluated. Results: In obesity, APO had an ameliorative effect on metabolic parameters, OS, inflammation, ED, programmed cell death and oxLDL uptake potential, but not on foam cell formation and LDL uptake potential. Conclusion: Apocynin may improve ED and AS in obesity by suppressing OS-linked factors involved in the early stage of AS.
