Article Biocompatibility Assessment of Chlorhexidine Gluconate Versus a Natural Mouthwash Containing Olea Europaea and Opuntia Ficus-Indica in Zebrafish Embryos(2025) Ünal, İsmail; Emekli-Alturfan, Ebru; Cansız, Derya; Yalcinkaya, Sebnem Ercalik; Beler, Merih; Eğilmezer, Gizem; Karagöz, AtakanVarious chemical solutions are available for oral hygiene and care, but these chemical solutions also cause various side effects in living systems. Therefore, the trend towards the production of highly biocompatible, natural-based oral care products has increased. This study aimed to compare the biocompatibility profiles of a conventional chlorhexidine gluconate-based mouthwash and a natural formulation containing Olea europaea leaf and Opuntia ficus-indica extracts using the zebrafish embryo model. Zebrafish embryos were exposed to two concentrations (100 ppm and 1000 ppm) of each mouthwash for 72 hours post-fertilization. Developmental properties such as mortality, hatching rate, pericardial edema, and body length were evaluated. Biochemical analyses included oxidative stress parameters and acetylcholinesterase (AChE) activity. Chlorhexidine exposure resulted in increased embryonic mortality, pericardial edema, and reduced body length. Biochemically, chlorhexidine increased lipid peroxidation and glutathione S-transferase (GST) activity while decreasing superoxide dismutase (SOD) and AChE activities. In contrast, embryos exposed to the natural formulation showed no significant developmental abnormalities and exhibited increased SOD and AChE activities without changes in lipid peroxidation. For this reason, vaccine studies and their urgent use are of great importance to prevent the pandemic. In this study, multi-organ damages caused by SARS-CoV-2 virus in human- angiotensin-converting enzyme type 2 (ACE2) transgenic mice and the protective effects of OZG-38.61.3 gamma irradiated SARS-CoV-2 vaccine against viral damage were investigated. Methods: For this purpose, transgenic K18-hACE2 BALB/c mice were randomly allocated into 4 groups, negative control group (NC), positive control group (PC, SARS-CoV-2 infected), and 2 different doses of OZG-38.61.3 vaccine (Challenge 1, dose of 10 13 and Ch2, 10 14 viral particle after SARS-CoV-2 infection). After the administrations, lung, heart and kidney tissues were examined by histopathological, immunohistochemical and TUNEL analysis. Results: Our results showed that the vaccine doses decreased the apoptosis, oxidative stress and inflammation parameters caused by virus in lung, heart, and kidney tissues. It was also found that the vaccine protected the expressions of tight junction proteins in the kidneys. Conclusion: According to our findings, it is suggested that the OZG-38.61.3 can be an effective and protective vaccine that can be safely used against the SARS-CoV-2 virus.Article Canlı Karaciğer Donör Adaylarında Hepatosteatozun Saptanmasında Biyopsi, Bilgisayarlı Tomografi ve Manyetik Rezonans Görüntülemenin Karşılaştırılması(2021) Sokmen, Bedriye Koyuncu; Şahin, Tolga; Koçak, Erdem; Oral, AlihanAmaç: Karaciğer donöründe hepatosteatoz (HS) varlığı, karaciğer transplantasyonu sonuçları üzerinde olumsuz etkilere sahiptir. Bu nedenle, donörde HS’nin tespiti, nakil öncesi dönemde hayati önem taşımaktadır. Bu çalışmanın amacı, canlı karaciğer donör adaylarında HS’nin saptanmasında karaciğer biyopsisi ve radyolojik yöntemlerin etkinliğini karşılaştırmaktır. Gereç ve Yöntem: Demiroğlu Bilim Üniversitesi’ne karaciğer transplantasyonu için donör adayı olarak kabul edilen 226 sağlıklı birey çalışmaya dahil edildi. Donörlerin demografik, histopatolojik, laboratuvar ve görüntüleme bulguları retrospektif olarak incelendi. Donörlerin bilgisayarlı tomografi (BT) ve manyetik rezonans görüntüleme (MRG) taramaları geriye dönük olarak yeniden değerlendirildi ve karaciğer yağ ölçümleri kaydedildi. Bulgular: Hastaların %39’u (88) kadın, %61’i (138) erkekti. Çalışma popülasyonunda ortalama yaş 34,3±8,7 yıl, ortalama ağırlık 78,0±12,6 kg, ortalama boy 169,1±9,6 cm ve ortalama vücut kitle indeksi 27,2±4,0 idi. Karaciğer biyopsisinde donörlerin %42’sinde <%5 HS vardı ve donörlerin %58’inde >%5 HS vardı. Hem BT hem de MRG, HS saptamada biyopsi ile anlamlı korelasyon gösterdi (p<0,05). Sonuç: Çalışmamızda MRG’nin BT kadar biyopsi ile ilişkili olduğu ve HS’nin saptanmasında rahatlıkla kullanılabileceği bulunmuştur. Karaciğer donörlerinde MRG kullanımı, iyonizan radyasyon içermemesinden dolayı nakil öncesi donör için daha uygun bir yöntem olabilir.Article Citation - WoS: 5Apocynin Ameliorates Testicular Toxicity in High-Fat Diet-Fed Rats by Regulating Oxidative Stress(Marmara Univ, inst Health Sciences, 2023) Hersek, Irem; Koroglu, M. Kutay; Coskunlu, Busra; Ertas, Busra; Sener, Goksel; Ercan, Feriha; Köroğlu, KutayObjective: The purpose of this study was to examine the effects of apocynin (APC), an inhibitor of NADPH oxidase (NOX), on high-fat diet (HF)induced testis cytotoxicity.Methods: Wistar albino rats were divided into three groups as control, HF and HF+APC groups. Rats in HF and HF+APC groups were fed using HF for 16 weeks and in the last four weeks of this period vehicle solution or APC (25 mg/kg) was administered orally five days a week, respectively. Control group was fed with standart lab chow for 16 weeks. Cholesterol, triglyceride, high-density lipoproteins, leptin, estrogen, testosterone, LH and FSH were estimated in blood serum. Sperm parameters were analysed from the epididymis. Testicular malondialdehyde, 8-hydroxy-2deoxyguanosine, glutathione, superoxide dismutase and myeloperoxidase levels were estimated biochemically. Testicular morphology, proliferative, apoptotic and NOX2-positive cells were analysed histologically.Results: HF-induced obesity caused significant alterations in serum lipid and hormone profiles. Testicular malondialdehyde, 8-hydroxy-2deoxyguanosine, and myeloperoxidase levels increased, glutathione and superoxide dismutase levels decreased in this group. Moreover, altered sperm parameters, increased degenerated seminiferous tubules, apoptotic and NOX2 - positive cells and decreased proliferative cells were observed in the HF group. All these biochemical and histological alterations improved in the HF+APC group.Conclusion: HF-induced obesity causes altreations in lipid values, sperm parameters and testicular morphology by increasing oxidative stress through NOX2 activity. Apocynin might prevent testis damage via regulating oxidant/antioxidant balance.Review The Use of Probiotics in the Treatment and Prophylaxis of Various Diseases in Turkiye: a Systematic Review(Bilimsel Tip Yayinevi, 2023) Kilbas, Elmas Pinar Kahraman; Altindis, MustafaThe aim of this study is to analyze the available information on the use of probiotics containing different microorganisms, singly or in combination, in order to investigate the effect of probiotics on the treatment, prophylaxis and microbiota of various diseases. In addition to Pubmed, Google Scholar, SCOPUS, EMBASE, Cochrane Library of Systematic Reviews, CINAHL, AMED, Turkish Medline electronic da- tabases, the gray literature was searched to cover the years 2004-2021. Randomized controlled studies (RCTs) included in the specified selection criteria were brought together. RCTs were evaluated using the Cochrane risk of bias tool. Of the 32 RCTs included in total, only yeast (Saccharomyces spp.) was administered in fourteen, only Lactobacillus spp. in six, Bifidobacterium spp. only in four, and probiotic supplementation containing more than one bacterial strain in six. Only 10/32 of the included studies fully reported the probiotic strain, and 22/32 reported only at the species level. In all nine studies using probiotics for the treatment of diarrhea, probiotics were reported to shorten the duration of diarrhea and length of hospital stay. Four of the five studies reporting the use of probiotics in the treatment of necrotizing enterocolitis reported that probiotics were not effective in reducing the incidence of necrotizing enterocolitis, but could improve nutritional tolerance. In two of the four RCTs investigating the use of probiotics in the treatment of Helicobacter pylori infec- tion, it was stated that the side effects associated with eradication therapy decreased. In both studies conducted for prophylaxis, it was reported that probiotics prevent antibiotic-associated diarrhea. This study shows that probiotics have positive results for the treatment indications and prophylaxis of various diseases. Guniz; Tiber, Pinar MegaBackground and Aims: The incidence of breast cancer is increasing day by day, especially in women. The search for new drugs against breast cancer is the focus of attention in research. Breast cancer and prostate cancer have remarkable biological similarities. Therefore, the 4-(4-chlorophenyl)-3-(1-(2-fluoro-[1,1'-biphenyl]-4-yl)ethyl)-5-((4-fluorobenzyl)thio)-4H-1,2,4-triazole (SGK597) compound that is suppressing cell proliferation in prostate cancer, was studied in MCF-7 breast can-cer and MCF-10A mammary epithelial cell lines. Methods: The WST-8 method was used to determine cell viability and cytotoxicity of SGK597 in MCF-7 and MCF10-A cell lines. The JC-1 test was applied to determine changes in mitochondrial membrane potential. The protein expression levels of Bax, Bcl-2, and c-PARP associated with apoptosis were determined using Western blot analysis.Results: After 24 and 48 hours of incubation of SGK597, the IC50 values were 28.74 pM and 17.28 pM for MCF-7; 65.9 pM and 50.5 pM for MCF-10A, respectively. Mitochondrial membrane potential showed a tendency toward depolarization in MCF-7 cells as a result of increasing concentration of SGK597, while the same tendency was not seen for MCF-10A. As a result of western blot experiments, no increase in the Bax/Bcl-2 ratio and c-PARP expression level was observed, indicating no apoptosis.Conclusion: It was observed that the compound SGK597 suppressed MCF-7 cell proliferation. These results indicate that SGK597 may be a candidate compound for use as an anticancer agent. However, the significant side effects of MTX limits its use. In this study, we aim to assess the hepatoprotective properties of Momordica charantia (MC) against MTX-induced liver damaged in rats.MATERIALS AND METHODS: Following one dose of MTX (20 mg/kg), the rats were given either distilled water or MC extract (300 mg/kg, po) for 5 days. After the dissection of the rats, the liver was removed to analyse tumour necrosis factor -a (TNF-a), interleukin-113 (IL-113), transforming growth factor 13 (TGF-13) and 8-hydroxy-2'-deoxy-guanosine (8-OhdG) levels and superoxide dismutase (SOD), catalase (CAT), and caspase-3 activities. The tissues were also examined histopathologically.RESULTS: The hepatic TNF-a, IL-113, TGF-13, 8-OhdG levels, and Caspase-3 activity in the MTX group were found to be significantly increased compared to the control group. However, MC extract was able to significantly decrease TNF-a, TGF-13, 8-OhdG levels, and Caspase-3 activity. Also, both the SOD and CAT activity of the MTX group decreased compared to the control group. Although only the SOD levels elevated significantly with MC treatment, the SOD and CAT activities of the MC treated group were similar to the control group. Supporting these biochemical parameters, MTX-induced histologic alterations in the liver were also ameliorated via MC treatment.CONCLUSION: Our results demonstrated that MC has a protective role against MTX-induced hepatic tissue injury by reducing apoptosis, oxidative damage, and the expression of pro-inflammatory cytokines.Review Molecular Characterization of Resistance and Virulence Genes in Enterococcus Faecium Strains Isolated Between 2000-2021; Systematic Review(Bilimsel Tip Yayinevi, 2022) Kahraman Kilbas, Elmas Pinar; Kilbas, Imdat; Ciftci, Ihsan Hakki; Kılbaş, Elmas Pınar KahramanIntroduction: The spread of Vancomycin-Resistant Enterococci (VRE) is a major threat in healthcare institutions, especially for patients in the risk group. The aim of this study is to reveal the antibiotic resistance genes, virulence genes and other accompanying factors detected in vancomycin resistant Enterococcus faecium strains isolated from various clinical specimens in different parts of Turkiye. Material and Methods: For this purpose, a systematic search was carried out using different electronic databases between January 2000 and September 2021. A total of 17 studies were evaluated within the scope of systematic review. Results: The vanA gene was detected the most between the years 2000-2007, and no statistically significant difference was found according to the years. The prevalence of the vanB gene was highest between 2008 and 2013, and no statistical difference was found according to the years (p> 0.05). The vanA gene was mostly detected in Eastern Anatolia, Black Sea, Mediterranean and Aegean, vanB Central Anatolia and Southeastern Anatolia regions. No reports related to the vanC gene were found. Since all strains were E. faecium in our study, it is an expected finding that the vanC gene region was never reported. The esp and hyl gene between 2014-2021. Conclusion: The prevalence of resistance and virulence genes among bacteria is a matter of great concern, limiting treatment options. In particular, effective measures should be taken to prevent healthcare-associated VRE infections, and each institution should report its own resistance data.
