    Bougainvillea glabra çiçeklerinden elde edilen ekstrenin fitokimyasal analizi ve in vivo Alzheimer hayvan modelinde etkisinin araştırılması

    Alzheimer's (AD) is one of the most common types of dementia and is a progressive neurodegenerative disease. AD is characterized by intracellular neurofibrillary tangles consisting of the hyperphosphorylated form of the neuron's microtubule-associated tau proteins, accumulation of toxic Aβ peptides, neuroinflammation, oxidative stress, and cholinergic conduction disorder (Ferrari et al., 2014). Aβ fragments produced by hydrolysis of amyloid precursor protein (APP) are thought to cause neuronal dysfunction and neuronal cell death. Oxidative stress contributes to the production of free oxygen radicals and subsequent neuronal cell death. (Chen and Zhong 2014). Moreover, chronic activation of microglia promotes neuronal damage through the production of proinflammatory cytokines. Recent data suggest that neuroinflammation may have a role in plaque deposition that begins before amyloid deposition and progresses (Kinney et al., 2018; von Bernhardi et al., 2015). Amyloid-targeted therapies have formed the basis of research in the field of AD until the last decade. However, increasing studies have shown that AD is dependent on more than one factor, and in line with these results, the search for drugs in the treatment of AD has shifted from an amyloid-centered treatment to a multi-targeted approach (Ferrari et al., 2014). The role of the cholinergic system in the pathophysiology of Alzheimer's disease has initiated the use of drugs acting on the acetylcholine molecule in the treatment of Alzheimer's, and the most successful of these drugs so far has been acetylcholinesterase inhibitor drugs. Today, synthetic, and herbal acetylcholinesterase inhibitor drugs are used in the treatment of Alzheimer's. Today, Physostigmine, Rivastigmine and Galantamine as herbal inhibitors are used in the treatment of Alzheimer's in the world. Huperzine A, whose research has come to an end in recent years, is an important indicator of plants in this regard (Gupta and Gupta, 1997; Feitosa et al., 2011; Ved et al., 1997). There are striking studies showing that the compounds detected in the plant Bougainvillea glabra have in vitro acetylcholine esterase inhibitor, anti-inflammatory, antiviral and anticancer effects. In vivo, the anti-inflammatory effect has been studied. (Bolognesi et al., 1997; Elumalai et al., 2012; Saleem et al., 2021) There is only one study for the treatment of AD of Bougainvillea species, and this study was conducted with the decoction of B. Spectabilis species (Abdel-Selam et al. 2021). There is no study showing the effect of B. glabra in the treatment of AD. Although some studies have shown that the Bougainvillea plant may have the potential to be a new drug for the treatment of neurodegenerative diseases (Abdel-Selam et al. 2017, Abdel-Selam et al. 2021), studies to support these findings have been limited and have not yet turned into a drug with an approved indication and used in the treatment. The subject of the project, which was designed and presented in the light of this information, is to investigate the effects of compounds obtained from Bougaivillea glabra, a plant grown in Turkey, in in vitro and in vivo Alzheimer's model. Within the scope of the project, Bougainvillea glabra plant will be collected from Muğla/Bodrum location, isolation, purification, and structural characterization of the active ingredient will be carried out. After ingredient analyzes, the efficiency of the extract in in vitro and subsequently in vivo Alzheimer's model will be examined. Before starting the in vivo study, single-dose/repeated-dose toxicity studies of the extract will be conducted. In in vivo examinations, both the processes developing in the brain tissue (inflammation, amyloid plaque accumulation, oxidative damage, etc.) that play a role in the formation of the disease, and the cognitive functions that occur due to the disease will be evaluated by receptor analysis and cognitive tests Alzheimer hastalığı (AH) en yaygın demans türlerinden biri olup progresif nörodejeneratif bir hastalıktır. AH’nın tarihi 1907'de başlamış, ancak patolojik nedenleri ve genetik bileşenleri yüzyılın sonuna doğru keşfedilmiştir. AH, nöronun mikrotübül ile ilişkili tau proteinlerin hiperfosforile formundan oluşan hücre içi nörofibriler yumaklar, toksik Aβ peptidlerin birikimi, nöroinflamasyon, oksidatif stres ve kolinerjik ileti bozukluğu ile karakterizedir (Ferrari vd., 2014). Amiloid öncül protein (APP)’nin hidrolizi ile üretilen Aβ fragmanlarının nöronal fonksiyon bozukluğuna ve nöronal hücre ölüme yol açtığı düşünülmektedir. Oksidatif stres ise serbest oksijen radikallerinin üretimine ve ardından nöronal hücre ölümüne katkıda bulunmaktadır. (Chen ve Zhong 2014). Ayrıca, mikroglia’nın kronik aktivasyonu, proinflamatuar sitokinlerin üretimi yoluyla nöronal hasarı ilerletir. Son veriler, nöroinflamasyonun amiloid birikiminden önce başlayıp ilerleyen süreçte plak birikiminde bir rolü olabileceğini düşündürmektedir (Kinney vd., 2018; von Bernhardi vd., 2015). Amiloid hedefli tedaviler son on yıla kadar AH alanındaki araştırmaların temelini oluşturmuştur. Ancak, artan çalışmalar, AH’nın birden fazla faktöre bağlı olduğunu göstermiş ve bu sonuçlar doğrultusunda günümüzde AH tedavisindeki ilaç arayışlarında, amiloid merkezli bir tedaviden çok hedefli bir yaklaşıma geçilmiştir (Ferrari vd., 2014). Alzheimer hastalığının patofizyolojisinde kolinerjik sistemin rolü, asetilkolin molekülü üzerinden etkili ilaçların Alzheimer tedavisinde kullanımını başlatmış ve bu ilaçlardan şimdiye kadar en fazla başarı elde edilenleri asetilkolinesteraz inhibitörü ilaçlar olmuştur. Günümüzde sentetik ve bitkisel kökenli asetilkolinesteraz inhibitörü ilaçlar Alzheimer tedavisinde kullanılmaktadır. Bugün bitkisel kökenli inhibitör olarak Fizostigmin, Rivastigmin ve Galantamin dünyada Alzheimer tedavisinde kullanılmaktadır. Son yıllarda araştırmaları bitim noktasına gelmiş Huperzin A da bitkilerin bu konuda önemli bir göstergesidir (Gupta ve Gupta, 1997; Feitosa vd., 2011; Ved vd., 1997) Bougainvillea glabra bitkisinde saptanan bileşiklerin, in vitro asetilkolin esteraz inhibitörü, antiinflamatuvar, antiviral ve antikanser etkilerinin olduğunu gösteren çarpıcı çalışmalar mevcuttur. In vivo olarak ise antiinflamatuar etki çalışılmıştır. (Bolognesi vd., 1997; Elumalai vd., 2012; Saleem vd., 2021) Bougainvillea türlerinin AH tedavisine yönelik tek bir çalışma mevcut olup söz konusu çalışma B. Spectabilis türünün dekoksiyonu ile yapılmıştır (Abdel-Selam vd. 2021). B. glabra türünün AH tedavisindeki etkisini gösteren bir çalışma ise mevcut değildir. Bazı çalışmalar, Bougainvillea bitkisinin nörodejeneratif hastalıkların tedavisi için yeni bir ilaç olma potansiyeline sahip olabileceğini gösterse (Abdel-Selam vd. 2017, Abdel-Selam vd. 2021) de bu bulguları destekleyecek çalışmalar sınırlı kalmıştır ve henüz onaylı endikasyonu olan ve tedavide kullanılan bir ilaca dönüşmemiştir. Bu bilgiler ışığında tasarlanan ve sunulan projenin konusu, Türkiye’de yetişen bir bitki olan Bougaivillea glabra’dan elde edilecek bileşiklerin in vitro ve in vivo Alzheimer modelinde etkisinin araştırılmasıdır. Proje kapsamında, Bougainvillea glabra bitkisi Muğla/Bodrum mevkiinden toplanacak, etken maddesinin izolasyon, saflaştırma ve yapı karakterizasyonu gerçekleştirilecektir. İçerik analizlerinden sonra ekstrenin in vitro ve takiben in vivo alzheimer modelinde etkinliği incelenecektir. İn vivo çalışma başlamadan öncelikle ekstrenin tek doz/tekrarlanan doz toksisite çalışmaları yürütülecektir. İn vivo incelemelerde ekstrenin gerek hastalığın oluşumunda rol oynayan beyin dokusunda gelişen süreçler (inflamasyon, amiloid plak birikimi, oksidatif hasar gibi.) ve gerekse hastalığa bağlı ortaya çıkan bilişsel fonksiyonlar reseptör analizi ve kognitif testler ile değerlendirilecektir.
